Dissolved oxygen can cause flow rate instability and increased baseline noise. Also, it has a quenching effect on fluorescence detection and increases the background of UV detectors. Dissolved gases can be released in the LC system, forming bubbles in check valves, at connections, or in detector flow cells.
In-line vacuum degassing is more effective at removing dissolved gas from mobile phases than sonication or helium sparging. In-line degassers work by drawing gas across a gas-permeable membrane encased in a sealed chamber. Traditionally, the membrane has been made of PTFE tubing, but the DEGASi PLUS degasser uses a highly permeable Systec AF membrane that is more gas permeable than PTFE. This translates into a 480 μL chamber volume for quick start-up and equilibration.
The DEGASi PLUS degasser employs the Systec ZHCR stepper-driven motor vacuum pump. Zero Hysteresis Constant Run (ZHCR) maintains a constant vacuum that is unaffected by varying degassing loads. This patented control strategy eliminates baseline fluctuation caused by dissolved gases and air.
Compatible with most HPLC solvents and buffers except hexafluoroisopropanol, hexanes (60% n-hexane), sodium azide, hydrofluoro and perfluorinated solvents, and fluorocarbons. Concentrated acids and bases, particularly 1 molar or greater, may attack PEEK and PPS wetted parts. Also, mobile phases with high concentrations of buffers and salts should not be left in an idle instrument. Flush system prior to shutdown to avoid crystallization.
